    Book SynopsisOver the last few decades, various techniques have been developed to alter the properties of plants and animals. While the targeted transfer of recombinant DNA into crop plants remains a valuable tool to achieve a desirable breeding outcome, integration of transgenes into the host genome has been random, which in part, leads to reduced acceptance of GMOs by the general population in some parts of the world. Likewise, methods of induced mutagenesis, such as TILLING, have the disadvantage that many mutations are induced per plant, which has to be removed again by expensive backcrossing. Advances in genome sequencing have provided more and more information on differences between susceptible and resistant varieties, which can now be directly targeted and modified using CRISPR/Cas9 technology. By selecting specific gRNAs occurrence of off-target modifications are comparatively low. ZFNs and TALENs- based approaches required re-engineering a new set of assembled polypeptides for every new target site for each experiment. The difficulty in cloning and protein engineering prevented these tools from being broadly adopted by the scientific community. Compared to these technologies, designing the CRISPR toolbox is much simpler and more flexible. CRISPR/Cas9 is versatile, less expensive and highly efficient. It has become the most widely used technology for genome editing in many organisms.Since its inception as a powerful genome-editing tool in late 2012, this breakthrough technology has completely changed how science is performed. The first few chapters in this book introduce the basic concept, design and implementation of CRISPR/Cas9 for different plant systems. They are followed by in-depth discussions on the legal and bio-safety issues accompanying commercialization and patenting of this emerging technology.     Book SynopsisThis book on ‘Secondary Agriculture’ discusses the goal of doubling farmers’ incomes. The term ‘secondary’ has a bearing on climate change adaptation and its mitigation, small farm viability and profitability, food security, nutrition, sustainable utilization of natural resources, and optimal usage of produce from primary agriculture and farm incomes. Promoting secondary agriculture has implications on attaining sustainable development goals, which aim to connect primary, secondary and tertiary sectors by using slack/idle factors of production, such as land and labour, contributing to primary agriculture production, capturing ‘value’ in primary agricultural activities, and generating additional income at the enterprise level. In context to same, the chapters of this book have been designed to promote secondary agriculture through low-cost skills and technology applications in agriculture and by upscaling knowledge via integrating primary, secondary and tertiary sectors of agriculture. The motivation behind this book is to address the challenges of biotic and abiotic stresses facing the farming community; to increase farmers income through low-cost skills and technology applications in agriculture; to upscale knowledge by integrating primary, secondary and tertiary sectors of agriculture. The food processing sector in India is still in a nascent stage with only 8 per cent of the produce being processed as against 80-98 per cent in case of high-income countries (Government of India, 2008, 2010). The food processing sector is now receiving the boost with the annual growth of 13.2 per cent in registered food processing units during 2004-10 (Government of India, 2011). Against this backdrop, there is a strong need to strategically handle the situation in order to facilitate a self-sustainable and long-run growth of the sector, which is felt possible by focusing on Secondary Agriculture. For these species, we describe morphology, ecogeographical distribution, phylogeny as well as cytogenetic and genomic features. For crops, we also address evolution under human selection, namely pre-domestication cultivation and domestication. We re-examine the genetic and archeological evidence of where, when, and how domestication occurred. We discuss unique aspects of genome evolution and maintenance under polyploidization, in natural and synthetic allopolyploids of the wheat group. Finally, we propose some thoughts on the future prospects of wheat improvement.     Book SynopsisThis open access book offers an introduction to mixed generalized linear models with applications to the biological sciences, basically approached from an applications perspective, without neglecting the rigor of the theory. For this reason, the theory that supports each of the studied methods is addressed and later - through examples - its application is illustrated. In addition, some of the assumptions and shortcomings of linear statistical models in general are also discussed. An alternative to analyse non-normal distributed response variables is the use of generalized linear models (GLM) to describe the response data with an exponential family distribution that perfectly fits the real response. Extending this idea to models with random effects allows the use of Generalized Linear Mixed Models (GLMMs). The use of these complex models was not computationally feasible until the recent past, when computational advances and improvements to statistical analysis programs allowed users to easily, quickly, and accurately apply GLMM to data sets. GLMMs have attracted considerable attention in recent years. The word "Generalized" refers to non-normal distributions for the response variable and the word "Mixed" refers to random effects, in addition to the fixed effects typical of analysis of variance (or regression). With the development of modern statistical packages such as Statistical Analysis System (SAS), R, ASReml, among others, a wide variety of statistical analyzes are available to a wider audience. However, to be able to handle and master more sophisticated models requires proper training and great responsibility on the part of the practitioner to understand how these advanced tools work. GMLM is an analysis methodology used in agriculture and biology that can accommodate complex correlation structures and types of response variables. Similarly, there is the need to produce the identified and highly commercially valued pharmaceutical compounds of mulberry under laboratory conditions through in vitro culture based secondary metabolite production through enhanced expression under the stress conditions or in presence of elicitors. Lately protocols have been developed for genetic transformation of mulberry through agrobacterium mediated and particle bombardment mediated gene transfer techniques. Biotechnology based molecular breeding techniques could also be utilized in raising the improved lines through marker assisted selection, soma clonal variations, mutational breeding, somatic hybridization, genome editing and other genetic engineering approaches. Apart from sericulture; mulberry should also be utilized and exploited in other sectors across the globe for additional revenue generation, for livestock maintenance, for environmental safety and in promoting human health. These diversified aspects of mulberry coupled with its economic importance in revenue generation through sericulture, animal husbandry and industrial products has prompted us to prepare this book. Two critical heat-sensitive stages in the lifecycle of the rice crop are targeted – the seedling and flowering stages – with screening based on simple phenotypic responses. The protocols are based on the use of a hydroponics system and/or pot experiments in a glasshouse in combination with a controlled growth chamber where the heat stress treatment is applied. The protocols are designed to be effective, simple, reproducible and user-friendly. The protocols will enable plant breeders to effectively reduce the number of plants from a few thousands to less than 100 candidate individual mutants or lines in a greenhouse/growth chamber, which can then be used for further testing and validation in the field conditions.
